DBscript: Sensitive and efficient

DBscript is sensitive and efficient with wide range of RNA input:To demonstrate the performance of DBscript Reverse Transcriptase in 2-step RT-qPCR setting, total RNA isolated from HEK cells was used with input amounts ranging from 1 µg to 1 pg. This corresponds to approximately 44,000 cells down to 0.04 cells or 4,400,000 RNA transcripts down to just 4.4 transcripts, assuming 100 copies of GAPDH RNA per cell. Three parallel sets of cDNA synthesis reactions were carried out with the following primers: Oligo(dT)20-VN (DB-1283; 2.5 µM), Random Hexamers (DB-1284; 1.25 µM), and a gene-specific primer for GAPDH detection (0.1 µM). Reactions were performed at 50 °C for 10 minutes, followed by termination at 94 °C for 2 minutes. Each reaction used 200 units of DBscript Reverse Transcriptase. Subsequently, 2 µl of cDNA were amplified in a PCR reaction using AptaTaq DNA Polymerase (DB-1277), primers, and a probe specific for GAPDH.
